چکیده

Erythropoiet ic cells from early chick embryos have been successfully ma in ta ined in vitro bo th in tissue culture (Murray, 1932; Settle, 1954; O'Brien, 1960; Spra t t and Haas, 1960 a and b; Hell, 1964; Levere and Granick, 1967; Wilt , 1967; Hagopian and Ingram, 1971; and Wenk, 1971) as well as in suspension culture (Hagopian et al., 1972). In all cases potent ia l erythropoiet ic cells will cont inue to differentiate into ma tu re erythrocytes dur ing the culture period. The present communica t ion described a technique which provides clean cultures of almost pure populat ions of erythroid cells from chick blastoderms. Some pre l iminary exper iments using the drugs ac t inomycin D and cordycepin are also reported.

Erythropoietic Cell Cultures from Chick Embryos

Erythropoietic cell cultures from very early chick blastoderms survive for several days They show four to seven doublings of the erythroid cells and the appropriate morphological changes from proerythroblasts to mature erythrocytes Cell cycle times are the same as in ovo for the first day of culture, but slow down thereafter The hemoglobins of both the primitive and the definitive red cell seri...

Developmental Changes of Erythropoiesis in Cultured Chick Blastoderms

The erythropoietic area of very early chick embryos was cultured as a tissue for up to nine days to study the changes in red cell type and hemoglobin type, the cell cycle time, the cell population kinetics, and the DNA synthetase activity of these cells. It was found that the area vasculosa without the participation of the embryo proper contained the information and the timing mechanism require...

Effect of microtubule inhibitors on the expansion of hypoblast and margin of overgrowth of chick blastoderms.

The effect of the microtubule inhibitors, Nocodazole and taxol, was studied on the expansion of fragments of chick hypoblast (8 to 10 h incubation) and of margin of overgrowth (24 h incubation) cultured on artificial substrata and on the epiboly in intact blastoderms (24 h incubation). Immunocytochemical staining of these cells with antiserum against tubulin showed that 1 micrograms Nocodazole/...

Behavioural transformation in chick yolk-sac cells.

During the first 3 to 4 days of development in the chick, the extraembryonic part, the yolksac, expands to encompass the yolk mass. The yolk-sac, initially a two-layered epithelial sheet, is pulled out by the action of a specialized band of cells at its periphery. These attach to the overlying vitelline membrane and move out over it. However, unincubated blastoderms are not attached to the vite...
